タイトル：Sleep spindles: a mechanistic biomarker for cognitive dysfunction in encephalopathy

概要：In epileptic encephalopathies, epileptic activity contributes to progressive cognitive dysfunction, but the mechanism is not well understood. Several epileptic encephalopathies share the trait of pathological spike-wave activation during non-rapid eye movement sleep (EE-SWAS), a state normally dominated by physiological sleep spindles, sleep oscillations known to coordinate offline memory consolidation. We find that sleep spindles predict IQ and sleep dependent memory in epilepsy, are disrupted by epileptic spikes that propagate to the thalamus, and can be increased by both pharmacologic interventions and non-invasive stimulation. Better understanding the circuits that support and disrupt these physiological and pathological oscillations will open opportunities to address cognitive symptoms in epilepsy and other neurologic and psychiatric disorders.
